Introduction to the principle and use of centrifuge
A centrifuge is a special instrument that separates and precipitates a mixed solution by centrifugal force.
First, the use of centrifuge:
1. Turn on the centrifuge power switch and enter the standby state.
2. Choose the right rotor: the liquid in the centrifuge tube should not exceed 2/3 of the total capacity when centrifuged , otherwise liquid, easy

2. Centrifuge tube balance error should be within 0.1 gram.
3. Select the centrifugal parameters:
a) Press the temperature setting button, then use the numeric keys to set the centrifugal temperature and press Enter to confirm.
b) Press the speed setting button to switch between RPM/RCF setting files and set the centrifugal speed with the number keys.
Enter to confirm.
c) Press the rotor setting button, then use the number keys to set the rotor model and press Enter to confirm.
d) Press the time setting button, then use the numeric keys to set the centrifugation time and press Enter to confirm.
e) Centrifuge brake or acceleration speed is generally set between 0 and 4 , and should not be adjusted frequently.
4. Place the balanced centrifuge tube symmetrically into the rotor. Cover the rotor cover and tighten the screws.
5. Press the centrifuge door. If the door is not closed, the centrifuge will not start.
6. Press the START button to start centrifugation. After the start of centrifugation, the centrifugal speed should be equal to the set speed.
Open, once the centrifuge is found to be abnormal ( such as unbalanced, causing the machine to vibrate significantly, or very noisy ) ,
Press the STOP button immediately . If necessary, directly press the power switch to turn off the power, stop the centrifugation, and find out the cause.
7. If the machine is found to be faulty, please contact the relevant personnel in time.
8. Please clean the rotor and centrifuge chamber after use, do not close the centrifuge cover, which will facilitate the evaporation of moisture.
9. You must register after use and indicate the usage.
Second, the use of centrifuges:
1. Set the operating temperature, usually 4 °C, first put the centrifugal turret into the centrifuge chamber, pay attention to the centrifugal shaft to clamp the shaft; close the door, the temperature of the centrifugal chamber begins to drop, and the pre-cooling time should be sufficient.
2. Load the sample into a suitable centrifuge tube, balance the weight with both balances, cover the centrifuge tube cap and tighten.
Note that the centrifuge tube is only 70% full, although it is covered, it may leak out due to too strong centrifugal force.
Most of the centrifuge tubes are covered with a lid, and the lid of the tube should be balanced.
The centrifuge tube is usually placed on crushed ice. When removing the balance, wipe off the ice-crushing liquid.
The centrifuge tube that is placed in the order should be equilibrated with another centrifuge tube filled with clean water.
3. Place the well-balanced centrifuge tube symmetrically into the centrifuge, cover the lid of the centrifuge, and pay attention to the tightening.
If the lid of the centrifuge is not tightened, it will fall out during centrifugation, causing great damage!
4. Close the centrifuge door and adjust the desired speed and time on the instrument panel; for example: 8000 rpm , 30 min .
Note that the speed used must not exceed the limit of the centrifuge. The maximum speed is usually written on the centrifuge. For example , the maximum speed of the TGL-20M is 20000 rpm . However, if the centrifuge is too old, it must be lowered. The speed and time settings should not be too high. For example, if you can use 7000 rpm, do not use 8000 rpm .
5. After confirming that all the steps are correct, press the Start button to start, and the centrifuge will gradually accelerate. At this time, it should be closely monitored.
Some old-fashioned time buttons, when set below 5min , must be turned 10mins and then turned back to 5min .
When pressing the Start button, the hold time should not be too short, please keep pressing for about one second before releasing.
It is the most dangerous when starting to accelerate. If you find that the sound is wrong or if there is a big shock, please press Stop immediately .
6. Wait until the centrifuge reaches the desired speed and make sure everything is normal before leaving.
7. When the centrifugation is completed, wait until the centrifuge is completely still before opening the hatch; please take out the tube as soon as possible, first observe whether the tube is completely, and the position of the sediment, pour out the supernatant as soon as possible, be careful not to get the sediment turbid.
Regardless of the supernatant or sedimentation, please use a clean beaker to collect the supernatant to avoid mistakes.
Be careful when dumping the supernatant so as not to pour out the sediment together; if you accidentally mix it together, you must come back again.
If there is a leak in the centrifuge tube, find out the cause and clean the centrifuge or centrifuge chamber immediately.
8. In the neutral between the two centrifugations, do not need to take out the centrifuge, but cover the door, do not allow hot air to flow into the centrifuge compartment.
9. After all the use, remove the centrifuge to clean, rinse with tap water, and put it upside down to dry. The centrifuge door should be opened. After the ice is melted, wipe it and clean it. If any liquid leaks out, wash it with water. Also check back the balance of the balance tube and the tabletop, it is easy to get dirty, and it must be carefully cleaned before leaving.
